Following graduation, he spent five years as an employee of the Monumenta Historica Germaniae. In 1903 he was appointed to a professorship in medieval history and historical auxiliary sciences at Ruprecht Karl University of Heidelberg. In 1924/25 he served as university rector. Hampe remained at Heidelberg until 1933 when he refused to cooperate with the increasing pressure put on universities by the new Nazi-led government of Germany and resigned his professorship.
